多杀菌素发酵工艺的优化及其毒性初步研究

摘  要:
对刺糖多孢菌S-6032发酵培养基及发酵工艺进行了优化,得到优化摇瓶发酵培养基为葡萄糖50g/L,麦芽糖10g/L,棉籽粉20g/L,硫酸铵1g/L,硫酸锌0.2g/L,玉米浆15g/L,牛肉膏2g/L,碳酸钙5g/L;确定摇瓶培养工艺为最佳接种量10%,最佳装液量为30mL/250mL摇瓶,最佳初始pH值为7.5。在优化培养条件下进行发酵培养,多杀菌素最终质量浓度达117.83mg/L,较优化前发酵培养条件下所得质量浓度(89.57mg/L)提高了31.6%。毒性实验表明发酵液对斜纹夜蛾幼虫毒杀效果显著,100mg/L多杀菌素的发酵液作用于斜纹夜蛾48h的致死率为90%。

摘  要:
Fermentation media and process for Saccharopolyspora spinosa S-6032 were optimized in the shake flask.The optimum media,which contained glucose 50 g/L,maltose 10 g/L,cottonseed powder 20 g/L,ammonium sulfate 1 g/L,zinc sulfate 0.2 g/L,corn steep liquor 15 g/L,beef extract 2 g/L and calcium carbonate 5 g/L,were obtained by orthogonal design experiment.And the optimum fermentation process,inoculum size of 10%,the liquid volume of 30 mL in the 250 mL shake flask and the initial pH of 7.5,were reached by single factor experiment.The ultimate production of 117.83 mg/L under the optimum conditions can be reached,which was increased by 31.6% compared to 89.57 mg/L on the original fermentation conditions.The toxicity test indicated that the broth with 100 mg/L spinosad had remarkable toxic effect on Prodenia liture(Fabricius).The lethal rate reached 90% after the pest was feeded with 100 mg/L spinosad for 48 h from the broth.
