鸢尾属部分种质资源遗传多样性的SRAP分析

摘  要:
利用SRAP分子标记对40份鸢尾属种质资源进行遗传特性及亲缘关系的分析研究,从170个SRAP随机引物中筛选出条带清晰、多态性高、重复性好且稳定性强的引物10个,共检测到168个位点且全部为多态位点,多态性条带比例为100%,表明供试材料具有丰富的遗传多态性。各种质之间的DICE遗传相似系数在0.067~0.740,平均相似系数为0.281。UPGMA聚类分析结果显示,当遗传系数为0.265时,可将供试材料分为7个类群。其中,19份德国鸢尾品种聚到一起,扁竹兰、喜盐鸢尾、德国鸢尾‘Queen’分别各自聚为一类,上海引种的马蔺和宁夏引种的马蔺聚为一类,而内蒙古马蔺则和路易斯安那鸢尾聚到一起,西伯利亚鸢尾、花菖蒲、黄菖蒲和3个德国鸢尾品种聚为一类,主坐标分析所反映的关系和UPGMA聚类图相似。

摘  要:
Sequence-related amplified polymorphism(SRAP) was used to detect the genetic diversity among 40 Iris L. materials. 10 primers which could produce clear, polymorphic and steady bands, screened out of 170 random primers, which were used and produced 168 bands totally. All of bands were polymorphic(100%), which indicated that the genetic diversity of Iris L. was rich. The genetic similarity coefficient among the accessions ranged from 0.067 to 0.740, and the average was0.281. At the point of the similarity coefficient 0.265, all the accessions might be separated into seven groups. 19 accessions of Iris germanica were brought together. Iris confusa Sealy, Iris halophila and Iris germanica ‘Queen' were classified separately. Iris lactea Pall. var. chinensis from Shanghai and Ningxia were lumped into the same category, while Iris lactea Pall.var. chinensis from Inner Mongolian and Iris louisiana were lumped together. Iris sibirica, Iris ensata, Iris pseudacorus and three accessions of Iris germanica were lumped into the same category. The results of principal coordinates showed similarity to the UPGMA dendrogram.
