香蕉对甘露糖和木糖的敏感性研究

摘  要:
以香蕉低代试管苗茎段薄层切片和香蕉吸芽薄层切片为外植体,分别设计了13个梯度,研究了香蕉在芽的再生和愈伤组织的诱导阶段对正筛选剂D-甘露糖和D-木糖的敏感性,为建立香蕉正筛选系统的遗传转化体系奠定基础。结果表明,(1)香蕉对D-木糖敏感,在D-木糖与蔗糖比例达到10∶20时,10 d开始显示出毒性,在D-木糖与蔗糖比例达到16∶14时能显著抑制香蕉芽的再生和愈伤组织的生长,当比例达到25∶5时完全抑制香蕉组织的生长,香蕉组织彻底死亡。因此,D-木糖能作为有效筛选剂对香蕉进行目的基因遗传转化;(2)香蕉对D-甘露糖不敏感,不适用于以D-甘露糖为筛选剂的遗传转化。

摘  要:
Genetic transformation is an important tool for banana improvement.However,as an economically important crop,consumed both as fruit and food,the safety of the genetically transformed banana should be carefully considered.Positive se-lection system,which utilize a key enzyme in metabolic pathway(such as xylA or pmi genes),is an effective way to solve the problem by avoiding antibiotic and other negative selection system,but these positive systems have not been used in banana transformation yet.This article describes the results of testing banana’s sensitivity to two positive selected agents,D-Mannose and D-Xylose,using banana thin cell layers derived from low-generation in vitro grown shoot tips as explants.13 concentra-tions of agents ranged from 0 to 30 g.L-1 were used and the results were shown as below:(1)Banana was highly sensitive to D-Xylose.Banana explants showed symptoms of toxicity at about 10 days after culturing on the medium containing D-Xylose and Sucrose at the ratio of 10∶20;The inhibition of the shoot and callus formation of explants were observed when the ratio was increased to 16∶14;and the explants completely died when the ratio was increased to 25∶5.Therefore,D-Xylose could be used as a selected agent in banana transformation.(2) Banana explants were not sensitive to D-Mannose and therefore D-Mannose was not a suitable agent for banana transformation.The banana’s sensitivity to D-Mannose and D-Xylose was dif-ferent.D-Xylose could be an effective agent for banana’s genetic transformation.The system is worth deep researching.
