紫锥菊提取工艺研究及有效成分含量测定

摘  要:
探索紫锥菊的乙醇法最佳提取工艺,并对菊苣酸含量进行测定。试验选取紫锥菊头状花序,用乙醇进行提取,采用L9(34)正交试验、以有效成分菊苣酸的含量为指标优选工艺参数。结果表明,紫锥菊的乙醇法最佳提取工艺条件为:料液比15倍量、乙醇浓度60%、提取温度90℃、提取1 h。以菊苣酸标准品为对照,以高效液相检测菊苣酸含量,色谱柱:Phenomenex C18110A(250 mm×4.6 mm,5μm),流动相:乙腈-0.1%磷酸水溶液(38∶62),流速:1.00 m L/min,紫外波长:326 nm,HPLC测定纯度为48.41%。此法提取工艺稳定、可行,测定方法简便准确,重复性好,可用于紫锥菊中菊苣酸的质量控制。

摘  要:
Objective: To explore the optimal ethanol extraction technology and determine the content of cichoric acid from Echinacea purpurea. Methods: Cichoric acid was extracted from Echinacea purpurea lead inflorescence site using ethanol and its content was determined by HPLC. The column of Phenomenex C18110A(250 mm×4.6 mm, 5 μm) was adopted with mixture of acetonitrile-0.1% phosphoric acid(38∶62) as mobile phase at flow rate of 1.00 m L/min, and the UV detector was set at 326 nm. The extraction process was optimized by L9(34) orthogonal design with cichoric acid as standard for quality control. Results:The best extraction condition obtained were as followed: the ratio of material to solvent was 1 ∶15, extracting for 1 hour each time by 60% of alcohol and 48.41% of purity was achieved by HPLC. Conclusion: This optimal technology is stable and workable. The determination method is simple, accurate and highly reproducible, so it can be used to control quality of cichoric acid in Echinacea purpurea.
