微波协同双水相提取桔梗茎总黄酮及抗氧化活性研究

摘  要:
利用微波协同双水相法研究了桔梗茎总黄酮的提取工艺及其体外抗氧化性。首先,采用单因素和正交设计法对提取工艺进行了优化;其次,利用分光光度法研究了桔梗茎总黄酮提取液对DPPH·、·OH的清除活性。结果表明,当料液比为1∶40(m/v),微波功率为500 W,醇水比为0.8∶1,提取时间为5 min,硫酸铵质量浓度为0.30 g/m L时,桔梗茎总黄酮的提取得率较高,其平均得率为0.793%;桔梗茎总黄酮提取液对DPPH·、·OH具有较强的清除能力,且其清除活性随桔梗茎总黄酮提取液质量浓度的增加而逐渐增强。

摘  要:
Aqueous two- phase cooperated with microwave methods were applied to explore the extraction technology of total flavonoids in Platycodon grandiflorum stems and its antioxidant ability in vitro was studied.First,the extraction technology was optimized by single factor and orthogonal test,while the scavenging activity of total flavonoids extraction solution to DPPH· and ·OH were measured by spectrophotometric method.When the ratio between material and solvent,the microwave power,the ratio between ethanol and water,the extraction time and the mass concentration of ammonium sulfate were 1 ∶ 40( m / v),500 W,0.8 ∶ 1,5 min and 0.30 g / m L respectively,the much higher average extraction yield of total flavonoids could reach about 0.793%.The activity test had shown that total flavonoids solution extracted from Platycodon grandiflorum stems had strong scavenging activity to DPPH· and ·OH,and the scavenging activity might gradually be enhanced with the increase of mass concentration of total flavonoids in extraction solution.
