HPLC-ELSD法测定甘草中单糖的含量

摘  要:
建立一种高效快速的高效液相色谱-蒸发光散射检测法(HPLC-ELSD)测定中药甘草中单糖———果糖及蔗糖含量的方法。采用CHROMATOREXNH2柱为固定相;以乙腈∶水为85∶15(V/V)为流动相;流速1.0mL/min;漂移管温度为84℃;氮气流速为2.5L/min。在上述色谱条件下,果糖和蔗糖的线性范围分别为1.422~3.318μg及12.028~60.140μg;且该二种单糖的检测限分别(信/噪=3)为4.74ng和3.31ng。该方法用于传统中药甘草中单糖的分离及含量测定,结果令人满意。

摘  要:
A simple and rapid method by HPLC with Evaporative Light Scattering Detection (ELSD) was developed for the determination of glucose, fructose and sucrose in traditional Chinese herb Glycyrrhiza uralensis Fisch. The HPLC separation was performed on a CHROMATOREX NH2 column using acetonitrile-water (85∶15, v/v) as mobile phase at a flow rate of 1mL/min. The drift tube temperature of ELSD was set at 84℃ with the nitrogen flow-rate of 2.2L/min. The linear ranges were from 1.422 to 3.318μg for fructose, and from 12.028 to 60.140μg for sucrose. The obtained detection limits (Signal/Noise=3) were 4.74ng and 3.31ng for fructose and sucrose, respectively. The determination result by this method was satisfactory in separation and determination of sugars in traditional Chinese herb G. uralensis Fisch.
