旋毛虫p43与p53核酸疫苗的构建及其免疫保护性

摘  要:
为探讨旋毛虫p43、p53基因的核酸疫苗对小鼠的免疫保护效果与免疫保护机制,将p43、p53基因分别克隆到真核表达载体pcDNA3.1(+)上,分3次肌肉注射免疫3组昆明小鼠。于三免后2周经口感染旋毛虫肌幼虫(100蚴/只),在攻虫后第7天和第35天剖杀小鼠,计算成虫减虫率、肌幼虫减虫率和繁殖力指数,评估其免疫效果。在各时间点上取小鼠血清用于检测抗体水平和脂肪酸结合蛋白(H-FABP)。结果显示,试验各组成虫减虫率与对照组相比差异显著(P<0.05),LPG及RCI减虫率与对照组相比均差异极显著(P<0.01);IgG抗体水平提高,与PBS组差异显著(P<0.05);各组H-FABP检测结果均低于70pg/mL。结果表明旋毛虫p43与p53基因的核酸疫苗具有较好的抗旋毛虫的免疫保护效果,且两核酸疫苗对小鼠心肌无损伤。

摘  要:
The aim of this study was to investigate the immunoprotection against Trichinella spiralis by DNA vaccines on the basis of p43 and p53 genes in mice.The DNA vaccines were constructed via cloning T.spiralis p43 and p53 genes into the eukaryotic expression vector pcDNA3.1(+),respectively. Intramuscular injection of the recombinant plasmids was performed in mice three times with two weeks interval. Two weeks after the third immunization,each mouse was perorally infected with 100 T.spiralis muscle larvae.To access the immunoprotectivity,the reduction rates of adult worms and muscle larvae and the index of reproductive capacity were calculated on the 7th day and the 35th day after inoculation.In addition,sera from mice were prepared for the detection of antibody levels and mouse heart-type fatty acid binding protein(H-FABP) at these two time points.The results indicated that a significantly higher antibody level was found in immunization group than in PBS group,and empty plasmid group.The IgG antibody level was improved and significant difference with the PBS group was observed.H-FABP test in each group showed that the index was lower than 70pg/mL.The DNA vaccines were proved to be safe for mice myocardium.
