考马斯亮蓝法测定茶籽多糖中蛋白质含量条件的优化

摘  要:
为建立高效、快速测定茶籽多糖中蛋白质含量的方法,以吸光度为考察指标,通过单因素和正交试验,对采用考马斯亮蓝法测定茶籽多糖溶液中蛋白质含量的试验条件(考马斯亮蓝G-250用量、反应时间、反应温度)进行了优化。结果表明,优化后的方法为:精确吸取茶籽多糖溶液1.0mL,加入考马斯亮蓝G-250溶液(0.01%)4.0mL,以蒸馏水补至总体积为10.0mL,混匀,25℃条件下放置35min,于595nm处测定其吸光度。该方法的精密度RSD为0.506%;稳定性RSD为0.836%;重现性RSD为1.57%;样品的平均加标回收率为99.19%,RSD为0.86%,表明该方法是测定茶籽多糖溶液中蛋白质含量的有效方法。

摘  要:
To establish a highly efficient determination method of protein content in Camellia oleifera seed polysaccharides,three factors including the amount of Coomassie brilliant blue G-250,reaction temperature and reaction time were optimized by single factor and orthogonal experiments using the absorbance as index.The optimized method was that 1.0mL solution of Camellia oleiferaseed polysaccharides and 4.0mL solution of Coomassie brilliant blue G-250were added together,and then the total volume of the solution reached 10.0mL by adding H2O.The solution was kept 35min at 25℃.The absorbency of the solution was measured at 595nm.The RSDof precision,stability and reproducibility was 0.506%,0.836% and 1.57%,respectively.The average recovery was 99.19%and the RSD was 0.86%.The method was effective for the determination of protein content in polysaccharides fromCamellia oleifera seeds.
