笃斯越橘CBF基因的克隆及序列分析

摘  要:
根据近缘植物中同源CBF基因的序列保守区设计引物,采用PCR方法,克隆出野生种笃斯越橘的CBF基因片段,将其克隆到pMD18-T Vector上。序列分析结果表明,笃斯越橘的CBF基因序列全长678 bp,编码225个氨基酸。同源性分析表明该基因的核苷酸序列和推导的氨基酸序列与其他植物CBF类基因具有较高的同源性,与桃DREB基因和蓝莓(Blue crop)DREB1基因氨基酸同源性高达85%以上。

摘  要:
Primers were designed according to the sequence of CBF gene in other plants.Fragment of CBF gene was isolated from Blueberry(Vaccinium uliginosuml L.) by PCR,and transferred into the pMD18-T Vector.The result derived from the sequence analysis showed that the sequences obtained the full-length cDNA was 678 bp long,and encoded a putative protein of 225 amino acids.Homology analysis showed that the nucleotide and the deduced amino acids sequences were homologous to the CBF genes in other species,especially strongly homologous to the DREB gene of Prunus persica and DREB1 gene of Blueberry(Blue crop) and the homology was up to above 85%.
