摘  要:
试验对1株H9N2亚型禽流感病毒血凝素(HA)基因进行扩增、克隆,然后对毒株的HA基因进行阳性克隆鉴定。测序结果表明:所扩增的HA基因全长1 683 bp,编码560个氨基酸;该毒株的裂解位点的氨基酸序列为R-S-S-R,属于低致病力毒株,该毒株有7个潜在糖基化位点,受体结合位点的氨基酸分别为YWT-NVLY,左侧壁氨基酸为NGQQG,右侧壁为GTSKA。将该毒株与国内一些参考毒株的HA基因序列进行比较分析,结果表明,核苷酸与氨基酸同源率较高,核苷酸同源率为93.0%~97.3%,氨基酸同源率为95.4%~96.6%,亲源关系比较近;与韩国毒株核苷酸和氨基酸同源率分别为83.3%和87.3%,亲源关系比较远。

摘  要:
The cDNA of HA gene of H9N2 avian influenza isolate was amplified,cloned and then sequenced.The result of its sequence analysis shows that cDNA of HA gene is composed of(1 683 bp) nucleotides,encoding for 560 amino acids.The sequence of the amino acids is R-S-S-R,typical of lowly pathogenic avian influenza.There are 7 potential glycosylation sites in the influenza virus isolate,the amino acids of the receptor-binding site are YWTNVLY,the amino acids of the left edge of receptor-binding site are NGQQG,and the amino acids of the right edge of receptor-binding site are GTSKA.The results of the comparative sequence analysis indicated 93.0%~97.3% of nucleotide homology and(95.4%~)96.6% of amino acid homology between the isolated H9N2 and the other from Chain and 83.3% of nucleotide homology and 87.3% of amino acid homology between the isolated H9N2 and the other from Korea.
