摘 要:
为探讨复方苦芩3种有效活性成分对人工感染肠炎型犬细小病毒病(canine parvovirus disease,CPVD)病犬小肠组织的保护作用,将50只试验犬随机分为复方苦芩总多糖组、复方苦芩总生物碱组、复方苦芩总黄酮组、空白对照组和模型组。其中,试验药物组均按1.5 mL/kg肌肉注射复方苦芩3种有效成分,2次/d,连续用药7 d后灌服2 mL(5×10~5 PFU/mL)的病毒液进行人工复制病理模型,空白组肌注射生理盐水,模型组只攻毒不给药。接种病毒后经检测为阳性后3天在无菌条件下取各组犬只小肠组织,用TUNEL法检测各组小肠组织细胞的凋亡率,同时采用荧光定量PCR对各试验组小肠组织Bax和Bcl-2 mRNA转录进行相对定量检测,并运用Western blot方法检测Bcl-2和Bax蛋白表达量。结果显示,通过组织切片观察,复方苦芩3种有效活性成分可防止病毒对犬小肠细胞形态的病理作用,可降低细胞凋亡率,并且3种有效成分对小肠组织可增高小肠组织Bcl-2/Bax基因和蛋白表达量,从而减少因病毒所引起的细胞凋亡;结果表明,复方苦芩3种有效成分对CPVD病犬小肠组织均有一定的保护作用,其中复方苦芩总生物碱对CPVD病犬小肠组织细胞凋亡的预防效果最显著。

摘 要:
To study the protective effects of 3 effective ingredients(total polyphenols,the total alkaloids and total flavonoid) of Kuqin compound on canine small intestine tissue in artificial intestinal infections with canine enteritis-type parvovirus,50 dogs were randomly divided into 5 groups,the total polyphenols group,the total alkaloids group,total flavonoid group,blank control group and model group.Those 3 effective ingredients groups were intramuscularly injected with 3 effective components of Kuqin compound at 1.5 mL/kg( body weight),2 times daily for 7 days,and 2 mL(5×10~5 pfu/mL) of the virus solution was artificially administered to replicate the pathological model.Those dogs in the blank group were injected with normal saline,and the dogs in model group was only injected with virus.The intestinal tissues of the dogs in each group were taken aseptically at 3 days after inoculating the virus,and the apoptotic rate of the cells in the canine small intestine of each group was detected by TUNEL method.At the same time,the transcription of Bax and Bcl-2 mRNA in small intestine tissues were detected by fluorescent quantitative PCR.And the protein expression of Bcl-2 and Bax were detected by Western blot.The results showed that,those 3 effective ingredients of Kuqin compound could reduce the apoptosis rate by preventing the virus from affecting the canine small intestine structural changes and increasing the Bcl-2/Bax gene and protein expression levels in the canine small intestine.Thus,we can make a conclusion that those 3 effective components of Kuqin compound have certain protective effects on parvovirus infected intestinal tissues,particularly,the total alkaloids of Kuqin compound have the most significant effect on the prevention of cell apoptosis in small intestine infected with canine parvovirus.
