摘 要:
目的利用硫酸卡那霉素(Kanamycin sulfate,KM)与呋塞米(furosemide,Fur)快速建立CBA小鼠急性耳毒性动物模型,观察给药时间对致聋的影响.方法 1、利用KM+Fur联合用药快速建立急性耳毒性动物模型:60只5~8周龄的健康CBA小鼠(雌雄各半)随机平均分成6组:正常组(空白对照)、KM+Fur联合用药后1 d组、用药后3 d组、用药后7 d组、用药后14 d组、用药后28 d组;2、在KM+Fur用药前和用药后1、3、7、14、28 d不同时间段检测运用听性脑干反应测试(auditory brainstem response,ABR)进行听力阈值及ABR波Ⅱ振幅分析;3、各组小鼠通过耳蜗基底膜HE染色铺片显微镜下观察小鼠耳蜗基底膜病理切片中毛细胞缺失和排列情况.结果 1、CBA小鼠联合运用KM+Fur后各时间点的ABR阈值与给药前相比,听力阈值明显升高,差异具有统计学意义(P<0.01);2、CBA小鼠联合运用KM+Fur后各时间点的ABR波Ⅱ振幅值与给药前相比较,幅度降低明显,差异具有统计学意义(P<0.01)3、CBA小鼠联合运用KM+Fur 1 d后,外毛细胞出现轻微的紊乱,随着损伤时间的延长,耳蜗外毛细胞的排列越来越紊乱;4、CBA小鼠联合运用KM+Fur后在不同时间段的ABR听力阈值及ABR波Ⅱ振幅值比较,差异均无统计学意义(P>0.05).结论硫酸卡那霉素和呋塞米联合一次给药,可以快速建立理想的CBA小鼠急性耳毒性模型.

摘 要:
Objective To establish an acute ototoxic model of CBA mice with Kanamycin sulfate(KM) and furosemide(Fur)and observe the effect of administration time on deafness. Methods First, Acute ototoxicity animal models were established rapidly by KM + Fur combination: Sixty healthy CBA mice(male and female) aged 5-8 weeks were randomly divided into6 groups: normal group(blank control), KM + Fur combined with drug 1 d group, 3 d after administration, 7 d after drug treatment group, 14 d after drug treatment group, 28 d after drug treatment group; Second, KM + Fur medication before 1,3, 7, 14, 28 d different time periods. The auditory brainstem response(ABR) was used to detect hearing threshold and amplitude of ABR wave Ⅱ. Third, The basement membrane of the cochlea in each group were stained by HE staining to observe the changes of the cochlear basal membrane Hair loss and alignment. Results The results showed that the ABR threshold at each time point after KM + Fur administration in CBA mice was significantly higher than that before administration(P<0.01); The combined use of CBA mice amplitudes of ABR II at each time point after KM+Fur were significantly lower than those before administration(P<0.01). After combined use of KM+Fur for 1 day, the outer hair cells appeared mild disturbance, with the extension of injury time, more and more cochlear outer hair cells disorder arranged;CBA mice combined KM + Fur at different time periods, the difference of ABR hearing threshold and amplitude ABR wave II was no statistical significance(P>0.05). Conclusion The combination of KM and Fur in one administration can rapidly establish the ideal acute ototoxicity model in CBA mice.
