镰形扇头蜱中2个新基因的分离、克隆与序列分析

摘  要:
为了对镰形扇头蜱中的生物活性分子进行相关研究,从中得到有价值的抗蜱候选分子,本试验从已构建的镰形扇头蜱半饱血雌蜱唾液腺cDNA文库中,随机克隆得到100个EST(expressed sequence tag)序列。其中8号和2号序列带有完整的po lyA尾,根据此2序列设计引物,经5′RACE(rap id am p lification of cDNA ends)得到2序列的全长基因,分别命名为RhHp 1和RhHp 2。RhHp 1基因全长1 823 bp,ORF(开放阅读框)为1 521 bp,编码区从110~1 631bp,编码506个氨基酸。RhHp 2基因全长1 002 bp,ORF为848 bp,编码区从31~879 bp,编码282个氨基酸。经蛋白序列分析,RhHp 1基因预期编码的蛋白与G enB ank中所登陆的序列同源性均小于20%,RhHp 2与其他序列同源性最高可达50.81%。经RT(reverse transcription)-PCR分析,与其他各发育阶段相比,RhHp 1基因在镰形扇头蜱卵中表达量较低;RhHp 2基因在镰形扇头蜱若蜱中低度表达,在其他各发育阶段表达量较高。结果表明,RhHp 1和RhHp 2基因为镰形扇头蜱唾液腺中分离出来的2个新基因,其表达量随着蜱的发育阶段而发生改变。此2个新基因所编码蛋白的结构和具体的分子生物学功能有待深入研究。

摘  要:
In order to get valuable anti-tick molecules,100 expressed sequence tags have been obtained from the cDNA library of salivary gland of feeding female Rhipicephalus haemaphysaloides haemaphysaloides,in which No.8 and No.2 had poly A sequences and were selected to study.5′ repid amplification of cDNA ENDS(RACE) and primers designed based on the sequences above were used to clone the two full length genes named as RhHp1 and RhHp2.The RhHp1 gene contained(1 823) bp and the coding region is from 110 bp to(1 631) bp((1 521) bp),encoding 506 amino acid residues;the RhHp2 gene contained(1 002) bp and the coding region is from 31 bp to 879 bp(848 bp),encoding 282 amino acid residues.Comparison and analysis of the deduced amino acid sequences were performed and the result showed that the highest homology of RhHp1 deduced amino acid with other sequences in GenBank was below 20%,and that of RhHp2 was 50.81%.Expression analysis by RT-PCR revealed that RhHp1 was lowly expressed in eggs when compared to other stages,RhHp2 was highly expressed in all stages of the tick except for the unfed nymphae.The study suggested that RhHp1 and RhHp2 were new genes isolated from salivary gland of the Rhipicephalus haemaphysaloides haemaphysaloides.In the different periods of tick development,the expression of RhHp1 and RhHp2 changed.The encoded proteins′ structure and molecular function of the two genes were under study.
