粗山羊草JAZ基因家族的全基因组鉴定与分析

摘  要:
为了挖掘可用于小麦茉莉酸调控通路及其穗部或花器官发育研究的候选基因,基于已发布的粗山羊草全基因组数据,利用生物信息学分析方法鉴定了粗山羊草JAZ基因家族,并从染色体分布、基因及蛋白结构、启动子顺式作用元件、系统进化关系和基因表达模式等方面对该基因家族的特征进行了比较分析。结果表明,本研究共鉴定到9个粗山羊JAZ基因,命名为AetJAZ1-AetJAZ9。染色体定位结果显示,9个粗山羊草JAZ基因分别定位在2D、5D、6D和7D染色体上,其中在7D染色体上分布最多。进化分析表明,粗山羊草JAZ蛋白与短柄草JAZ蛋白亲缘关系最近,且9个粗山羊草JAZ蛋白被分别聚类到了S5、S12、S13、S16和S20五个亚族中,其中S20亚族有五个粗山羊草JAZ蛋白。启动子分析表明,9个粗山羊草JAZ基因启动子顺式作用元件种类和数量存在差异,推测其各自的功能也存在差异。表达谱分析表明,粗山羊草JAZ基因在粗山羊草不同组织间存在不同的表达特性,且不存在组成型表达基因,其中,AetJAZ1和AetJAZ2表现出明显的组织特异性,分别只在穗部、雌蕊和雄蕊中特异表达。

摘  要:
In order to eveal the candidate genes for the regulation pathway of jasmonic acid and the development of ear or floral organs in wheat,based on the available genome data of Aegilopstauschii,the JAZgene family was identified through bioinformatics analysis in this study.The chromosome distribution,gene and protein structures,cis-regulatory elements of promoter,phylogenetic relationship,and gene expression patterns of this family were comprehensively compared.Results showed that a to-tal of nine JAZgenes in Aegilops tauschii genome were found and then designated as AetJAZ1-AetJAZ9.The result of chromosomal localization showed that nine AetJAZgenes were located on chromosome 2D,5D,6Dand 7D,respectively.High density of AetJAZgenes were observed on chromosome7 D.Evolutionary analysis showed that AetJAZ proteins and JAZ proteins from Brachypodium distachyon shared closer affinities,and the AetJAZ gene family was divided into S5,S12,S13,S16 and S20subgroups.A total of five AetJAZ proteins were clustered into S20 subgroup.The promoter analysis indicated that the species and quantities of cis-elements in nine AetJAZgenes promoters had some differences,which suggested each AetJAZgene had different functions.The expression profiles analysis demonstrated that each AetJAZgene had different expression patterns among different tissues in Aegilops tauschii.None of nine AetJAZgenes was constitutive expression gene.AetJAZ1 and AetJAZ2 exclusively expressed in spike,pistil and stamen,respectively.
