葡萄根癌病病原菌类型鉴定及几种杀菌剂和抗菌素对其抑菌效果

摘  要:
以‘巨峰’葡萄茎部及根部癌瘤为材料,于培养基上分离根癌病原菌相似菌株,采用与生长素合成有关基因序列引物及葡萄土壤杆菌6b特异基因序列引物对这些菌株DNA进行PCR扩增,其中有6株菌株中扩增获得特异性目的条带,鉴定为葡萄土壤杆菌章鱼碱类型。通过对向日葵的接种试验,筛选出毒性最强株系。采用抑菌圈法检测了9种杀菌剂和抗菌素对该菌株的抑菌效果。结果表明:72%农用硫酸链霉素对该菌株的抑制作用显著,其200倍稀释液抑菌圈直径为38.84mm;其次为三氯异氰尿酸和80%乙蒜素,二者抑菌圈直径分别为17.37mm和10.66mm。参试的20%龙克菌、80%多菌灵、3%中生菌素、40%可杀得三千、2%春雷霉素、25%叶枯唑对该菌株无明显抑制作用。

摘  要:
The bacteria collected from crown gall that located on the basal part of the stems and on the roots in 'Kyoho' grapevines were cultured on medium in petri dishes.Those strains similar to the Agrobacterium were selected for the biotype identification.PCR analysis was carried out for the strains by using the primers that related to the synthesis of auxin and to the 6b specific gene in Agrobacterium.Six of the strains showed the typical bands,which could be classified to the biotype of octopine.The strain with the strongest pathogenicity was screened out by inoculated into the stem of the sunflower seedlings.Then,the bacteriostasis of nine bactericides or antibiotics was tested to this strain by using the inhibition zone method.The results showed that 72% streptomycin sulfate for agricultural use grade with the 200 times dilution had a significant inhibition to the growth of the strain with a diameter of inhibition zone of 38.84 mm,followed by trichloroisocyanuric acid and 80% ethylicin with the diameter of inhibition zone of 17.37 mm and 10.66 mm,respectively.The rest ones including 20% thiodiazole-copper,80% carbendazim,3% zhongshengmycin,40% cocide,2% kasugamycin,and 25% bismerthlazol showed no significant effect on the inhibition.
