梨果皮色泽变异品种(系)的AFLP与SRAP标记鉴定

摘  要:
为揭示梨果皮色泽变异品种(系)与原始品种的遗传差异,利用AFLP和SRAP标记对3组果皮色泽的变异品种(系)进行了鉴别。54对AFLP引物共检测1 089个位点,其中多态性条带数为688,多态性比率为63.2%;20对SRAP引物可检测210个位点,其中多态性条带为76条,多态性比率为36.2%。分析结果表明,7对AFLP引物、3对SRAP引物可区分南果与其红色变异系;3对AFLP引物、1对SRAP引物可区分考密斯、红考密斯、早红考密斯与其绿色变异系;所检测的引物均无法区分红安久及其绿色变异系。综合AFLP与SRAP标记的检测结果,获得4个品种(系)的特征谱带,SRAP引物em11/me5可区分除红安久及其变异系以外的所有供试材料,可应用于各品种(系)的分子鉴别。

摘  要:
In order to reveal the genetic difference between the skin colour mutants and their original varieties,molecular identification and analysis were conducted on three groups of skin colour variations in pear by AFLP and SRAP makers.A total of 1 089 gene loci were screened by 54 AFLP primer-pairs,of which 688 were polymorphic with the polymorphism percentage of 63.2%.And 210 gene loci were screened by 20 SRAP primer pairs,of which 76 were polymorphic with the polymorphism percentage of 36.2%.Results showed that seven AFLP and three SRAP primer pairs could distinguish Nanguo and red variant strain;three AFLP and one SRAP primer pairs could identify the difference among Doyenne du Comice,Red Doyenne du Comice,Early Red Doyenne du Comice,and green variant strain of Early Red Doyenne du Comice;Red D Anjou and green variant strain could not be distinguished by any AFLP and SRAP primer pairs.Combining with data revealed by AFLP and SRAP,specific patterns of 4 cultivars or strains were constructed.All cultivars or strains,except Red D'Anjou and its variant strain,could be identified by SRAP primer em11/me5,and this primer could be used as a useful marker in molecular identification.
