解淀粉芽孢杆菌B10-26对芝麻的促生防病效果及其定殖能力分析

摘  要:
为了确定解淀粉芽孢杆菌B10-26的生防效果与定殖能力,采用浸种法、灌根法、利福平标记法、PCR法等对其促生作用、防病效果、定殖能力、胞外酶活性、抗生素合成酶相关基因进行检测。结果表明,1×10~8cfu/mL的B10-26发酵液对芝麻种子的萌发率及芝麻幼苗的根长、株高和鲜、干质量都有显著促进作用,其中幼苗干质量增加46. 51%; B10-26发酵液与菜豆壳球孢同时接种芝麻幼苗时,防治效果为37. 2%,而在B10-26发酵液灌根7 d后再接种菜豆壳球孢,防治效果达到61. 1%; B10-26菌株具有植株内部定殖能力,在芝麻植株根部定殖量最高,播种后12 d时达到6. 1×10~4cfu/g;胞外水解酶活性检测结果表明,B10-26菌株可以产生β-1,3-葡聚糖酶、蛋白酶、纤维素酶,不产几丁质酶;对B10-26菌株基因组中抗生素合成相关基因进行检测发现,其具有脂肽类抗生素合成相关基因fenB和srf AD以及聚酮类抗生素合成相关基因baeB。综上,解淀粉芽孢杆菌B10-26具有良好的促生防病及定殖能力,有较好的实际应用价值。

摘  要:
In order to determine the biocontrol effect and colonization ability of Bacillus amyloliquefaciens B10-26,the plant growth promoting function,control effect,colonization capacity,secretion capacity of extracellular enzyme and genes related to antibiotic biosynthesis were detected through seed-immersion method,root drenching method,rifampicin notation method and PCR method,respectively. The results showed that 1 × 10~8cfu/mL B10-26 fermentation liquid significantly promoted the germination rate of sesame seeds,root length,plant height and fresh,dry weights of sesame seedlings,especially that the dry weight of sesame seedlings increased by 46. 51%. And the control effect of B10-26 on sesame stem rot was 37. 2% when bacterial suspension of B10-26 and mycelium suspension of Macrophomina phaseolina were inoculated simultaneously. However,the effect reached 61. 1% when mycelium suspension of Macrophomina phaseolina was inoculated after 7 days of inoculation of bacterial suspension of B10-26. Thestrain B10-26 was able to colonize within the sesame seedlings,and the colonization of B10-26 was the highest in root,reaching 6. 1 × 10~4cfu/g at 12 days. The result of enzyme secretion test showed that the strain B10-26 was able to produce β-1,3-glucanase,protease and cellulase,while the activity of chitinase was not detected in dish. When the antibiotic synthesis-related genes in B10-26 genome were amplified,the genes fen B and srf AD of lipopeptide antibiotic synthase and the gene baeB of polyketide antibiotic synthase were detected. Due to its excellent capacity of growth-promoting,biocontrol and colonization,the strain B10-26 has good practical application value.
