玉米ZmbZIP基因植物表达载体的构建

摘  要:
根据玉米(Zea mays)ZmbZIP的基因序列和植物表达载体pCAMBIA3301的多克隆位点设计带有限制性内切酶位点的特异性引物,以质粒pGM-T-ZmbZIP为模板PCR扩增ZmbZIP基因片段,双酶切目的片段及载体,回收后连接,构建该基因的植物表达载体。结果表明,扩增出的ZmbZIP基因片段长度为894 bp。经PCR检测及测序鉴定,表明植物表达载体构建成功,为进一步研究该基因的功能奠定了基础。

摘  要:
According to the restriction enzyme sites of expression vector pCAMBIA3301 and sequence of ZmbZIP gene,a pair of primers containing restriction enzyme sites were designed.The ZmbZIP gene was obtained by PCR using pGM-T-ZmbZIP as templet.PCR product and the plasmid pCAMBIA3301 were digested by the corresponding restricted enzymes respectively,and then the ZmbZIP gene was cloned into pCAMBIA3301 vector.The results showed that the fragment length of ZmbZIP gene was 894 bp.PCR and sequencing results suggested that plant expression vector of ZmbZIP gene was constructed successfully,which provided an effective tool for the further study of ZmbZIP gene function.
