苏云金芽胞杆菌晶体毒素对体外培养的日本血吸虫成虫的毒力

摘  要：
将人工感染日本血吸虫尾蚴 45d的家兔主动脉灌注冲虫获得成虫 ,经Earle’s液洗涤 ,加入细胞培养板内 ,用含 10 %牛血清的RPMI 16 40培养液于 37℃、5 %CO2 培养箱中培养 ,苏云金杆菌B 0 0 1、E 0 0 2提取的伴胞晶体毒素 ,以不同浓度加入到各孔中。 2 4h后成虫运动缓慢 ,并有部分死亡 ,随着伴胞晶体毒素作用时间的延长 ,成虫的死亡率也增加。当毒素总蛋白含量为 117.2 9μg/ml时 ,经菌株B 0 0 1晶体毒素作用的成虫死亡率在2 4h时为 87.74% ,48h时为 10 0 % ,LD50 分别为 2 1.42 μg/ml、9.77μg/ml;经菌株E 0 0 2晶体毒素作用的成虫死亡率在 2 4h时为 86 .18% ,在 48h时为 10 0 % ,LD50 分别为 33 .16 μg/ml、13.19μg/ml。这表明两种苏云金杆菌晶体毒素均对血吸虫成虫有杀伤作用。

摘  要：
Adult Schistosoma japonicum was obtained 45 days after rabbits were infected with Schistisoma japonicum cercaria from the skin. The adults were washed by Earle's solution and placed into hole of cell culture plate. Bacillus thuringiensis B?001,E?002 crystal toxin was with different concentrations added in every hole of cell plate. The adults were cultivated in RPIM1640 medium containing 10% bovine serum in 5% CO\-2 incubator at 37℃. Twenty four hours later the adults moved slowly and some died. The deuth rate increased with increase in the duration of toxic treatment. The death rate of adults treated with B?001 crystal toxin at a dose of 117.29 μg total protein/ml were 87.74 % and 100% for 24 h and 48 h respectively. The LD 50 values were 21.42 μg and 9.77 μg total protein/ml respectively. The death rate of imagoes treated with for E?002 crystal toxin were 86.18% and 100% for 24 h and 48 h respectively. The LD 50 values were 33.16 μg and 13.19 μg total protein/ml respectively. This study indicated that the parasporal crystal toxin of Bt B?001, E?002 is highly toxic to adult Schistosoma japonicum .
