不同途径免疫小鼠重组乳酸乳球菌诱导的黏膜及系统免疫应答分析

摘  要:
重组乳酸乳球菌分别经鼻腔和口腔免疫小鼠后,免疫后的测定时间内鼻腔免疫方法刺激呼吸道和肠道产生s-Ig A水平高于口服免疫方法,且肺泡冲洗液中s-Ig A水平差异极显著(P>0.01),肠道黏膜s-Ig A水平无显著差异(P>0.05);血清中Ig G抗体水平两种免疫方法均无显著差异(P>0.05),但鼻腔免疫组抗体水平在最后一次免疫后7 d达到最高,在整个测定时间内维持在高水平,口服免疫组抗体水平在最后一次免疫后21 d达到最高水平,然后迅速下降;h1型细胞因子IL-2和IFN-γ水平滴鼻免疫组高于口服免疫试验组,差异不显著(P>0.05),Th2型细胞因子IL-4水平在各组之间差异不显著(P>0.05)。结果表明,重组乳酸乳球菌分别经鼻腔及口腔免疫小鼠后,能诱导机体产生黏膜免疫系统的体液免疫及Th1型细胞免疫反应,且滴鼻免疫方法优于口服免疫方法,为传染病的预防控制提供了新的免疫策略。

摘  要:
With the aim of determining the mucosal immune pathways of Recombinant Lactococcus lactis,we observed that Re-combinant Lactococcus lactis administered intranasally and orally respectively induced mucosal and systemic immune responses in mice.At the whole experimental session, the production of the s-Ig A antibody in lung lavage fluid from the i.n immunized mice was significantlyhigher than that from the o.r immunized mice(P<0.01)the production of the s-Ig A antibody in intestinal washes from the i.n immunizedmice was higher than the o.r immunized mice(P<0.05). Ig G anti-PRRSV was detected in serum,the level of special Ig G from the i.nimmunized mice was higher than the o.r immunized mice(P<0.05),however,at all time points. the i.n immunized mice reachedbn itsmaximum number on day 30,which lasted at least 4 weeks post-immunization;as well as the o.r immunized mice get it's maximum numberon day 44,following decreased rapidly;Interestingly,we investigated the activity of Th1-type cytokines IL-2,IFN-γ and Th2-type cytokinesIL-4 in splenocytes on day 14 after the last immunization,the contents of cytokines IL-2 and IFN-γ in intranasal immunized mice washigher compared to that from the oral immunized mice(P>0.05),Notably,contents of IL-4 cytokine was not significantly different betweendifferent groups. All the results indicated that mice immunized by the recombinant L.lactis expressing ORF6 gene of PRRSV couldsignificantly induce mucosal immune responses,humoral immunity responses and enhanced Thl immunity response against the PRRSV,in contrast,Intranasal immunization is superior to oral immunization.
