摘  要:
为研究Prdx1在AH(Aniline Hydrochloride)诱导溶血性贫血小鼠模型中调控巨噬细胞清除脾脏沉积血红素的作用及其相关分子变化,腹腔注射AH,采用倒置显微镜、流式细胞仪、动物血液分析仪检测小鼠的溶血性贫血症状;通过免疫组织化学、Western Blot方法和RT-PCR技术检测Prdx1调控巨噬细胞清除脾脏沉积血红素的相关信号分子变化。结果表明,AH通过抑制HO-1的翻译水平诱导的Prdx1-/-小鼠脾脏血红素沉积显著高于野生型小鼠,HO-1的mRNA水平显著提高从而负反馈调节Nrf2的转录水平,引起Nrf-2/HO-1信号通路阻滞。

摘  要:
To investigate the role and related molecular changes of Prdx1 eliminated spleen deposited heme through regulating macrophages in the mice model of hemolytic anemia induced by Aniline Hydrochloride,after the intraperitoneal injection of AH,used inverted Microscope,Flow Cytometer and Animal Hematology Analyzer to observe the hemolytic anemia of mice. Through immunohistochemistry,Western Blot method and RT-PCR technique,the expression and transcriptional level of protein which related with macrophage removes spleen depositional heme regulated by Prdx1 was detected. The results showed that AH induced Prdx1-/-mice spleen heme deposition was significantly higher than that of the wild-type mice by inhibition of HO-1 translation level. The level of HO-1 mRNA significantly increased,thus negative feedback regulated Nrf2 transcription caused the Nrf-2/HO-1 signaling pathway block.
