Expression and thermal stability analysis of Peat1 and the 3 deletion mutants.

摘  要:
A research was conducted to reveal the functions of nascent polypeptide-associated complex (NAC) and ubiquitin-associated (UBA) domains in the thermal stability of Peat1 (plant activator protein from Alternaria tenuissima). Fusion expression vectors of Peat1 protein and the 3 deletion mutants were constructed. The recombinant plasmids were induced by IPTG and the target proteins (Peat1, Peat1- Delta CD99, Peat1- Delta ND49 and Peat1- Delta ND108) were expressed, obtained by AKTA and their thermal stability was analysed. The results revealed that 3 deletion mutants had good thermal stability like Peat1. The research demonstrated that the coexistence of NAC or UBA domains is not necessary for the thermal stability of Peat1 protein, and they may separately give the protein a particular structure stability.
