摘  要:
家蚕丝腺拥有较强合成丝蛋白的能力,通过利用这一能力,使其能够大量生产有价值的蛋白,人们开始引入转基因家蚕,使它能够在丝腺中合成重组蛋白并分泌到蚕茧中。获得的重组蛋白可在生物学、生物技术,和药学领域中得到广泛应用。我们通过控制合成蛋白在丝腺中的定位,在后部丝腺(PSG)中表达重组蛋白使其定位在不能溶解的丝素中,在中部丝腺(MSG)中表达则定位于亲水外层的丝胶层中。本文主要讨论将转基因家蚕丝腺生物反应器作为优良工具,应用于大量生产药用蛋白和在工业化水平生产相关重组蛋白的可能性,以及生产重组蛋白过程中所面临的挑战和未来的应用前景。

摘  要:
The silk gland of silkworm Bombyx mori has the excellent ability of synthesizing silk proteins. To utilize such capacity for mass production of useful proteins,transgenic silkworms were generated that synthesized recombinant proteins in the silk gland and secreted them into the silk cocoon. These recombinant proteins can be widely used in the biological,biotechnical and pharmaceutical applications. By controlling the expressed regions of the recombinant protein in the silk gland,Expression in the PSG or MSG led to localization in the insoluble fibroin core or hydrophilic outer sericin layer,respectively. This review focuses on using the silk gland of silkworm Bombyx mori as a bioreactor,and the possibility of utilizing it as a valuable tool for the mass production of therapeutic and industrially relevant recombinan proteins. The challenges and perspectives of recombinant protein production are also discussed.
