低聚壳聚糖激发子对杨树抗病性的诱导作用

摘  要:
以壳聚糖(脱乙酰几丁质)为原料,采用H2O2氧化法降解壳聚糖制备低聚壳聚糖激发子,通过诱导处理杨树愈伤组织,研究了低聚壳聚糖激发子对杨树抗病性的诱导作用。结果表明:低聚壳聚糖激发子对PAL、几丁质酶、β-N-乙酰葡萄糖苷酶、SOD活性具有明显的诱导作用,酶在达到各自的峰值时其活性是无菌水(对照)的2.83、3.35、2.78、4.8倍。酶活性的增高表明低聚壳聚糖激发子能够诱导杨树的抗病反应,提高抗病性。病原菌接种试验进一步证实了经激发子诱导处理后杨树愈伤组织抗病性增强,明显抑制病原菌菌丝的生长,降低感病指数。在一定浓度范围内,激发子的诱导作用随浓度增加而增强,浓度为10μg.mL-1时诱导效果达到最大,再增加浓度诱导,诱导作用不再增强。

摘  要:
The chitosan-oligosaccharides elicitor was made with H2O2 to decompose chitosan,and induction of the elicitor to disease resistance of poplar callus was studied.The result showed that chitosan-oligosaccharides elicitor could induce the notable increase of PAL,chitinase,β-N-acetyl-D-glucosaminidase,SOD activity,and the highest activity of the 4 enzymes were 2.83,3.35,2.78,4.8 times as high as the each control enzyme activity.Poplar callus treated with chitosan-oligosaccharides elicitor could notable restrain mycelium growth of Dothiorella gregaria,and reduce susceptible index.It indicated that the chitosan-oligosaccharides elicitor could start and speed up expression of the resistant gene.Accompanied with the fast expression of the gene,the resistant metabolism in the plant were increased,and the resistant products accumulated rapidly,the resistance of the poplar cell was increased.
