鄂梨2号叶片高效再生体系的建立

摘  要:
以早熟砂梨品种鄂梨2号组培苗叶片为外植体,建立叶片再生体系。结果表明,鄂梨2号叶片再生不定芽最佳培养基为NN69+1.0 mg/L TDZ+0.1 mg/L IBA,愈伤组织再生率达到100%,不定芽再生率为36.67%;不定芽增殖最佳培养基为MS+1.0 mg/L 6-BA+0.2 mg/L IBA,增殖系数为2.19;不定芽在1 000mg/L ABT中浸蘸5 s后接种在添加500 mg/L活性炭的1/2MS培养基上,生根率最高,为66.67%;试管苗移栽成活率为84.44%。

摘  要:
Leaves excised from Eli 2 Hao were used as explants to establish regeneration system. The results indicated that the maximum adventitious buds regeneration frequency achieved 36.67% on the mediu of NN69+1.0 mg/L TDZ+0.1 mg/L IBA.The most suitable medium for shoot multiplication was MS+1.0 mg/L 6-BA+0.2 mg/L IBA and the average multiplication coefficient was 2.19. The buds were dipped in 1 000 mg/L ABT for 5 s and inoculated on 1/2 MS medium with 500 mg/L activated carbon. The highest rooting rate was 66.67%, and the survival rate of plantlets was 84.44%.
