藏山羊FGF10基因克隆及其组织表达分析

摘  要:
为了探讨藏山羊成纤维细胞生长因子10(fibroblast growth factor 10,FGF10)基因分子结构特征,并阐明其在藏山羊不同器官组织中的表达情况,试验采用逆转录PCR(reverse transcription-PCR,RTPCR)克隆该基因,并采用实时荧光定量PCR(qPCR)技术检测FGF10基因在藏山羊各器官组织中的表达情况。结果表明:克隆获得藏山羊FGF10基因序列长度为1 252 bp,包括完整CDS区642 bp(Gen Bank登录号为KX530811),编码213个氨基酸,蛋白质分子质量为23.78 ku,属于疏水性不稳定蛋白,且主要分布于细胞外。与山羊FGF10基因CDS区比对结果显示,藏山羊FGF10基因序列存在一个核苷酸突变位点,并导致该位点缬氨酸(Val)突变为异亮氨酸(Ile)。组织表达结果显示,FGF10 mRNA主要在藏山羊肺脏、脾脏和背最长肌中高表达,而在心脏、肝脏和肾脏器官组织中表达水平较低。

摘  要:
To explore the molecular structural characteristics of fibroblast growth factor 10 gene( FGF10) in Tibetan goat and its expression levels in different tissues,RT-PCR was used to clone the gene in this study and qPCR was used to detect the mRNA expression levels of FGF10 in different tissues of Tibetan goat. The results showed that the full-length sequence of Tibetan goat FGF10 gene was 1 252 bp containing 642 bp in CDS region( Accession No. KX530811) which encoded 213 amino acids. The molecular weight of FGF10 protein was 23. 78 ku,which belonged to hydrophobic unstable protein and this protein was mainly distributed in the extracellular matrix. Sequence alignment showed that a nucleotide mutation existed in the CDS of Tibetan goat FGF10 compared with that in goat FGF10,which caused a Val-to-Ile mutation in protein expression. The tissue expression results showed that FGF10 mRNA was highly expressed in lung,spleen and longissimus dorsi tissues,but lowly expressed in heart,liver and kidney.
