摘  要:
[目的]为促进贵州白水牛资源保护与开发的科学性,同时为品种选育提供更加充分的科学依据。[方法]本实验以贵州水牛(灰色)为对照组,以贵州白水牛为实验组分别构建DNA池,对不同毛色贵州水牛的MC1R基因和TYR基因进行PCR扩增并确定SNP位点,并利用生物信息学软件分析突变位点对RNA的二级结构,蛋白质的二、三级结构的影响。[结果]在贵州水牛(灰色)和贵州白水牛扩增的MC1R基因中共同筛选到错义突变exon1-T843C,导致缬氨酸(Val)变成丙氨酸(Ala),还在贵州水牛(灰色)中筛选到exon1-C1228T(同义)、exon1-G1330A两个突变位点。在两种水牛扩增的TYR基因共同筛选到2个错义突变exon1-G826A、exon5-T70C,分别导精氨酸(Arg)突变为组氨酸(His)、亮氨酸(Leu)突变为异亮氨酸(Ile);1个同义突变exon3-A83G。

摘  要:
【Objective】To promote the scientific protection and exploitation of the buffalo resources in Guizhou,and to provide more scientific basis for breeding.【Method】In this experiment,the Guizhou grey water buffalo was used as the contrast group,while the Guizhou white water buffalo was used as the experimental group.All DNA samples from the above groups were used to construct the DNA pool,respectively.The MC1 Rand TYR genes of Guizhou buffalo with different hair color ware used to carry out PCR amplification and scanning single nucleotide polymorphisms(SNPs).Bioinformatics software analysis was used to analyze SNPs loci and their effect of RNA secondary structure and the secondary and tertiary structure of proteins.【Result】In the MC1 Rgene amplification of Guizhou gray buffalo and Guizhou White Buffalo,a missense mutation in exon 1(T843C),leading valine(Val)to alanine(Ala)was founded,as well as two mutations in exon 1(C1228T)(synonymous mutation)and exon1(G1330A)were screened in the buffalo(gray)in Guizhou.In two buffaloes,SNPs identification of TYR gene showed two missense mutations in exon 1(G826A)and exon5(T70C),respectively,which resulting arginine(ARG)to histidine(His),leucine(Leu)to isoleucine(ILE),respectively.Besides,a synonymous SNP in exon3(A83G)was founed.
