In order to reveal the influence of karst mountainous terrain on population distribution, the Puan county in Guizhou province was selected for research area. The elevation, slope, topographic relief were extracted by using the geographic information system and overlaid the graph of population in 2010 respectively. The results showed that with the elevation between 640 to 1 640 meters, population and population density increase with elevation while with the elevation between 1 640 to 2 100 meters, population and population density reduce as the elevation increases. It forms a unique phenomenon of vertical distribution of population. Under the influence of slope factor, population distribution reduces as the slope increases, and population density and slope have a negative correlation. Under the influence of topographic relief factor the population distribution increases when the topographic relief decreases. Topographic relief and population distribution also have an obvious negative correlation.
