春兰根状茎的增殖与分化条件优化

摘  要:
以春兰种子无菌萌发的根状茎为材料,采用正交设计方法,研究了春兰根状茎增殖与分化的适宜条件。结果表明,根状茎增殖适宜的培养基为1/2MS+3 mg·L-1 BA+蔗糖3%+香蕉5%+琼脂0.8%,根状茎分化适宜的培养基为1/2MS+3.0 mg·L-1 BA+1.0 mg·L-1 NAA+0.8 mg·L-1 IBA+蔗糖3%+琼脂0.8%。使用100 g·L-1香蕉作为附加物,春兰根状茎在培养30d后重量可达初重的2.1倍。根状茎在固体培养过程中不发生褐变现象,添加活性炭能促使根状茎产生根毛类似物。

摘  要:
Taking the rhizome from Cymbidium goeringii seeds germination as the materials,designing orthogonal experiments,the appropriate conditions of the C.goeringii rhizome multiplication and differentiation were studied.The results showed that the appropriate medium was 1/2MS +3 mg·L-1 BA+sucrose 3% + Banana 5%+ agar 0.8% for multiplication and 1/2MS +3.0 mg.L-1 BA+1.0 mg·L-1 NAA+0.8 mg·L-1 IBA + sucrose 3% + agar 0.8% for differentiation.Using the 100 g·L-1 banana juice as attachment could make the weight of rhizome be 2.1 times as the original weight after 30 days'culture.There was no browning phenomenon when the rhizome cultured in the solid medium.The addition of activated carbon to medium could promote the occurring of root hair analogues from rhizome.
