单诺沙星在鲫鱼体内的药物代谢动力学研究

摘  要:
以15 mg/kg的剂量肌肉注射单诺沙星,研究其在鲫鱼体内的药物动力学代谢规律。取给药后不同时间的鲫鱼血浆,采用反相高效液相色谱法测定血浆中单诺沙星的质量浓度,用MPTCP软件进行数据处理和分析。结果表明,健康鲫鱼肌注给药的药时数据符合二室开放模型,主要药物动力学参数为:表观分布容积(V)为5.099 0 L±0.061 5 L,分布半衰期(t1/2α)为0.527 5 h±0.008 1 h,消除半衰期(t1/2β)为286.944 0 h±18.595 0 h,药时曲线下面积(AUC)为387.767 7/(mg/L).h±23.779 3/(mg/L).h。单诺沙星在鲫鱼体内的主要药物动力学特征为分布快且完全,消除缓慢,作用时间长。

摘  要:
The plasma pharmacokinetics of danofloxacin were investigated in Crucian Carps after single intramuscular administration of the injection 15 mg/kg.The blood samples were collected at different time and danofloxacin concentrations were determined using reversed-phase high performance liquid chromatography(RP-HPLC).The data were treated by MCPKP pharmacokinetic software.The results indicated that plasma concentration-time data of danofloxacin mesylate were fitted to a two-compartment open model,with disposition and elimination half-life(T1/2α,T1/2β) is 0.527 5 h±0.008 1 h and 286.944 0 h±18.595 0 h;Apparent volume of distribution(V) is 5.099 0 L±0.061 5 L.Area under the plasma druge concentration-time curve(AUC) is 387.767 7/(mg/L)·h±23.779 3/(mg/L)·h;The main pharmacokinetic parameters were as follows:absorbed rapidly and completely,eliminated slowly and long effect time.
