新疆紫草秋水仙素诱变及倍性鉴定研究

摘  要:
【目的】探索秋水仙素诱变及倍性鉴定的技术条件。【方法】采用不同浓度的秋水仙素分别对新疆紫草幼苗茎尖和无菌苗茎尖进行点涂、振荡浸泡处理,采用形态学、气孔及染色体计数法对变异株进行鉴定。【结果】对新疆紫草幼苗采用点涂法,秋水仙素浓度为500 mg/L时,幼苗的成活率和变异率分别为67.44%、40.59%。其变异株叶片宽度明显大于对照株,变异株的气孔长度和宽度显著大于对照株,变异株叶片下表皮每个视野平均气孔数目(9个)较对照株(19个)明显减少;对新疆紫草无菌苗采用振荡浸泡法,秋水仙素浓度为700 mg/L、处理时间为4 h时,成活率和变异率分别为68.18%、86.67%;秋水仙素浓度在200~500 mg/L范围内,茎尖细胞染色体数目为28条的比例最高达68.4%;诱变后茎尖成活率还受培养基中激素组合的影响;秋水仙素诱变后产生大量形态变异。【结论】影响染色体加倍效果的因素是秋水仙素浓度和处理时间。

摘  要:
【Objective】The project aims to explore the technology conditions on colchicine mutagenesis and identification of ploidy.【Method】Arnebia Euchroma( Royle) Johnst seedlings and sterile seedlings stem tips were dot-daubed and shocked with different concentrations of colchicine respectively,and the variation strains were identified by morphology,porosity and chromosome counting methods. 【Result】Seedlings survival rate and variation rate reached 67. 44% and 40. 59% respectively when the dot-daubed concentration of colchicine was 500 mg / L. The variant strains leaves width were significantly bigger than those of the normal strains leaves width,and variant strains stomatal length and width were all significantly bigger than thase of normal strains. The average 9 number stomata of variant strains leaves hypodermis under each view was significantly fewer than that of control strains( 19); The Arnebia Euchroma( Royle) Johnst sterile seedlings survival rate and variation rate were 68. 18% and 86. 67% respectively when soaked with 700 mg / L colchicine and shocked on shaker for 4h; Stem cells chromosome number was 28( Fig 4c) and the highest proportion of 68. 4% when colchicine concentration ranged in 200 mg / L ~ 500 mg / L; Stem tips survival rate was also affected by the medium of hormone combination; Colchicine mutagenesis produced a large number of morphological variation.【Conclusion】The factors affecting chromosome doubling effect are colchicine concentration andtreatment time.
