1株溶磷细菌的筛选及其溶磷物质分析

摘  要:
为了获得高效溶磷菌并了解其溶磷机制,从生活垃圾堆积地采集土样,筛选出10株对卵磷脂和磷酸钙均有溶解能力的菌株,通过液体摇瓶复筛,得到1株高效兼溶磷酸钙和卵磷脂的溶磷菌LY8,培养6d后2种难溶磷发酵液中水溶性磷质量浓度分别达到647.8mg/L和26.6mg/L。结合生理生化特征和16SrDNA序列分析,初步鉴定其为巨大芽孢杆菌(Bacillus megaterium)。在不同难溶性磷源条件下LY8分泌的主要溶磷物质不同,对于无机难溶磷磷酸钙,LY8菌株分泌的主要溶磷物质是有机酸,其次是磷酸酶;而对于有机难溶磷卵磷脂,LY8菌株分泌的主要溶磷物质是磷酸酶,且其分泌的有机酸、蛋白质和多糖可能也具有一定的溶磷效果。

摘  要:
To obtain high-efficiency phosphate-solubilizing bacteria and understand the phosphatesolubilizing mechanism,ten bacteria strains which could solubilize tricalcium phosphate and lecithin were isolated from the soil in dump sites,and the strain LY8 was the most effective for solubilizing tricalcium phosphate and lecithin,the water-soluble phosphorus concentration in fermentation broth was up to 647.8mg/L and 26.6mg/L respectively,after cultivation for six days.After physiological characteristics identification and 16 Sribosomal DNA gene sequence analysis,strain LY8 was identified as Bacillus megaterium.Phosphate-solubilizing substances of extracellular secretion were different for various forms of insoluble-phosphate,which were mainly organic acids and phosphatase secondly for tricalcium phosphate.However,phosphatase was the most important for lecithin,and the organic acids,polysaccharides and proteins also had the effect of solubilizing phosphate probably.
