紫叶李花粉生活力及贮藏特性研究

摘  要:
[目的]探讨适宜紫叶李花粉萌发的最佳培养基和最佳贮藏条件。[方法]以新鲜紫叶李花粉为材料,以0、50、100、150、200和250mg/L的蔗糖浓度配制培养基,以0、10、20、30、40mg/L的浓度配制硼酸溶液,用染色法和培养基萌发法研究不同浓度的蔗糖和硼酸对花粉萌发的影响,并探讨不同贮藏条件对紫叶李花粉萌发的影响。[结果]紫叶李花粉不添加蔗糖和硼酸培养基的萌发率较低,仅为5.5%,随着蔗糖和硼酸浓度的升高,其萌发率呈现先升高后降低的趋势。最佳培养基为:蔗糖100mg/L+硼酸30mg/L,花粉萌发率可达26.7%,比对照提高3.9倍。贮藏花粉的最佳条件为-80℃,耐贮藏力长达30d,25d后萌发率达到8.8%。[结论]不同浓度的蔗糖和硼酸对紫叶李花粉萌发有显著影响;超低温是贮藏紫叶李花粉的最佳贮藏条件。

摘  要:
[Objective] The study aimed to discuss the optimum medium and the optimum storage condition for the pollen germination of Prunus ceraifera. [Method] With the fresh pollens of P. ceraifera as the tested material,the different media were prepared with the cane sugar at 0,50,100,150,200 and 250 mg/L and the boric acid solution was prepared with the boracic acid at 0,10,20,30 and 40 mg/L. The effects of the cane sugar and boron with different concn. on the pollen germination of P. ceraifera were studied by dyeing method and medium germination method and the effects of different storage condition on the pollen germination of P. ceraifera were also discussed. [Result] The germination rate of P. ceraifera pollens was lower on the medium without adding the cane sugar and boracic acid,only being 5.5% and it showed the trend of first ring and then decreasing along with the increase of the cane sugar and boracic acid concn. The optimum medium for the pollen germination was 100 mg/L sugar + 30 mg/L boric acid,which cloud get the pollen germination of 26.7%,being 3.9 times higher than that of CK. The optimum condition for the storage of pollen germination was -80 ℃,under which,the most storage time for the pollen was up to 30 d and the germination rate still reached 8.8% after storage for 25 d. [Conclusion] The cane sugar and boron with different concn. had the remarkable effects on the germination of pollen P. ceraifera and the super-low temperature was the optimum storage condition for the germination of P. ceraifera pollen.
