一种家蚕胚胎发育因子的分子克隆及分析

摘  要:
利用表达序列标签(Expression sequence tag,EST)和电子克隆等技术,克隆家蚕BmEDF基因cDNA电子序列,经RT-PCR生物验证正确,登录GenBank(DQ443161)。BmEDF基因cDNA长1098bp,ORF全长810bp,编码产生269个氨基酸。BmEDF基因编码的蛋白分子量约为30.7kDa,等电点(PI)为9.16。本研究将对今后分子水平分析胚胎发育提供研究基础。

摘  要:
Using a bioinformatics approach and expression sequence tag database, cDNA of the Bombyx mori embryonic development factor (BmEDF) was successfully acquired in silico. After verified by reverse transcript polymerase chain reaction (RT-PCR), this BmEDF was accepted by GenBank (Accession no. DQ443161). The whole cDNA sequence covers 2327bp, which contains a complete open reading fragment and encodes a 269 amino acid protein. M.W. and PI of the predicted BmEDF in Bombyx mori are 30.7 kDa and 9.16 respectively. This experiment provided us with a feasible bioinformatics method for cloning new functional genes, thereby preparing for further research on development of silkworm.
