小花棘豆中毒对家兔脑组织α-甘露糖苷酶的影响

摘  要:
探讨小花棘豆对家兔脑组织α-甘露糖苷酶(AMA)的影响,进一步揭示小花棘豆的毒性作用机理。将24只家兔随机分为4组,即对照组和试验Ⅰ、Ⅱ、Ⅲ组。将小花棘豆全草粉碎后,按试验Ⅰ组添加15%,试验Ⅱ组添加30%,试验Ⅲ组添加45%的比例制作混合饲料,饲喂至典型临床症状出现为止。攻毒后第14、35、70 d每次每组随机采集2只家兔的全脑,检测家兔不同脑区AMA活性及表达的变化。结果显示,小花棘豆中毒可不同程度地导致家兔脑组织AMA活性和表达下降,从第14d起,所有试验组小脑AMA活性及mRNA表达量均显著低于对照组(P﹤0.05),试验Ⅲ组家兔大脑和丘脑AMA活性及mRNA表达量均显著低于对照组(P﹤0.05),但3个试验组海马和脑干AMA活性及mRNA表达量与对照差异均不显著(P﹥0.05)。结果表明,不同剂量小花棘豆均可降低家兔脑组织AMA活性及其mRNA表达量,小脑、大脑和丘脑是其主要作用的靶区,而且小脑的变化较大脑和丘脑明显。

摘  要:
To study Oxytropis glabra DC on rabbit brain tissue α-mannosidase(AMA) effect,and further reveal the mechanism of the toxicity of O. glabra DC,24 rabbits were randomly and equally divided into 4 groups(the control group and experimental groupⅠ,Ⅱ,Ⅲ). The dried plant of O. glabra DC was comminnuted,and different amounts of the grass powder(15%,30% and 45%) were mixed with the feeds in the three experimental groups. The rabbits consumed the forages freely until typical symptoms were observed. 2rabbits were selected randomly from each group on the 14 th,35th and 70 th day respectively,the brain was collected and the contents of AMA and its mRNA expression in different encephalic regions were measured. The results demonstrated that O. glabra DC inhibited the content and expression of AMA,the content of AMA and the expression of AMA mRNA in cerebellum in all experimental groups were significantly lower than normal control group from 14 th day(P ﹤ 0. 05),the content of AMA and the expression of AMA mRNA in cerebrum and thalamus in experimental group Ⅲ were significantly lower than normal control group(P ﹤ 0. 05),but the difference of the content of AMA and the expression of AMA mRNA were not obvious in brainstem and hippocampus(P ﹥ 0. 05). The results showed that O. glabra DC has remarkable effects on the contents of AMA of brain in rabbits. It is indicated that three regions were the effect target sites of O. glabra DC in the brain tissue,we concluded that O. glabra DC could produce poisoning effect by promoting cerebellum,cerebrum and thalamus.
