桂林蛇源裂头蚴分离株的分子鉴定及种系发育关系分析

摘  要:
为研究从桂林草花蛇中分离到的裂头蚴的分子分类地位,对其核糖体18SrDNA、28SrDNA、ITS全基因及线粒体cox1部分基因进行了克隆和序列分析。从GenBank获取相应基因序列,应用CLUSTAL W2软件进行多重比对分析序列差异,应用MEGA 4.0软件构建种系发育树。结果表明,在基于18S、28S基因序列构建的种系发育树中,本株裂头蚴分别与欧猥迭宫绦虫(D64072)18S构成一个单独分支,自展值为100%;与拟曼迭宫绦虫(AF004718)、欧猥迭宫绦虫(AF004717)、无头蚴绦虫(AF096223)28S同属一个分支,自展值61%。基于ITS1、ITS2和cox1部分基因序列构建的种系发育树与所有欧猥迭宫绦虫均构成同一亚群,总分支自展值分别为100%、99%、64%,三者在不同的分离株之间呈现基因多态性,从总分支自展值来看cox1比ITS更适合用于种内遗传多态性研究,ITS适合做定种的分子标记。

摘  要:
To study the taxonomy position of plerocercoid from flower-waved snake in Guilin city,the nuclear 18 S rDNA,28 S rDNA,internal transcribed spacers(ITS) genes and the partial mitochondrial cyclooxygenase 1(cox1) gene of the plerocercoid were cloned,and the sequences analyzed.The corresponding sequences were downloaded from GenBank,and were aligned by the Clustal W2,and the phylogenetic trees were constructed by MEGA 4.0.The result showed that Guilin isolates and the S.erinaceieuropaei(D64072)18 S were clustered in the same clade based on the 18 S phylogenetic tree,bootstrap 100%.Guilin isolates were also in the same clade with S.mansonoides(AF004718)、S.erinaceieuropaei(AF004717),Sparganum sp.(AF096223) in 28 S,bootstrap 61%,and in the same group with all of S.erinaceieuropaei ITS1,ITS2 and partial mitochondrial cox1 gene sequences,the total branch bootstrap values were 100%,99%,64%.It showed gene polymorphism between different isolates in ITS and cox1,cox1 is more suitable than ITS in studying intra-specific genetic polymorphism which considered from the bootstrap values,and ITS could be used as genetic marker for the species identification.
