Effects of Different Culture Media on Seed Germination of Pleione bulbocodioides (Franch.) Rolfe

摘要：

[Objective] To discover the optimum culture condition for seed germination of Pleione bulbocodioides(Franch.) Rolfe and provide technical support for the acquisition of test-tube plantlets.[Method] Effects of sucrose concentrations,active carbon concentrations,basal medium types,4 hormones(6-BA,KT,NAA and IAA) concentration and 2 combinations of double-hormones on the germination of Pleione bulbocodioides(Franch.) Rolfe seeds that were obtained through artificial pollination were investigated in immature embryos.[Result] The optimal culture condition for seed germination was a combination of 1/2MS,2% sucrose,0.05% casein,0.75% active carbon,0.55% agar and 1.5 mg/L IAA(pH 5.8).The addictive IAA could cause a higher germination frequency,which induced more and larger protocorms whose average diameter reacheed 1-1.5 mm.[Conclusion] A good and effective seed germination system was established in this study,which would provide basis for acquisition of abundant test-tube plantlets of Pleione bulbocodioides(Franch.) Rolfe.
