多酚与淀粉样纤维相互作用新说

摘  要:
先前的研究均发现,多酚类小分子能够抑制低浓度的蛋白质淀粉样纤维的形成,或者是切断、改造淀粉样纤维。我们首次发现,在高浓度淀粉样纤维条件下,多酚类小分子化合物能够通过疏水作用、π-π共轭作用、氢键作用吸附到球蛋白淀粉样纤维的表面,驱动处于液晶态的纳米纤维超分子自组装,在空间形成饱含水分的三维立体网状多层次结构,即水凝胶。并且,这种相互作用不依赖于对蛋白质的特殊选择性。这种水凝胶中的淀粉样纤维通过自身的物理结构特性(电位、疏水性、构象等),诱导食源性致病菌凝聚,进而破坏其细胞结构,实现广谱的抗菌作用。同时,对正常人体细胞没有影响,所以安全可靠。

摘  要:
It was found in previous studies that one of the most impressive properties of polyphenols is the inhibition of amyloid protein fibril formation. In addition,they remodel or cut mature amyloid fibrils under low protein concentrations. We found for the first time that polyphenols-binding amyloid fibrils in the nematic phase(high protein concentration) induce controlled fibrils aggregation to form complex hierarchical structures with water-rich nature,so-called hydrogels without phase separation,which are mainly driven by the exogenous complementary hydrogen bonding,π-π stacking and hydrophobic interactions. Changing the type of amyloid does not significantly change the conclusions of the structure and properties of the hydrogels,which indicates that the effect of polyphenols on controlling gelation of amyloid fibrils is not protein-dependent. The ensued hydrogels exhibit strong antibacterial activities against both gram-negative and gram-positive bacteria,controlled by bacterial agglomeration,without any notable cytotoxicity to human cells. The anti-bacteria mechanism of the amyloid fibril hydrogels is initiated by membrane disintegration. The cationic,hydrophobic and conformational properties of the amyloid fibrils could be the main sources for the observed antibacterial activity.
