致病性迟缓爱德华氏菌CA26的分离鉴定及其对鲶鱼免疫因子的影响

摘  要:
采用平板划线培养的方法,从凤阳养殖场患病鲶鱼肝脏样本中分离纯化到病原菌菌株CA26,使用VITEK-32全自动细菌分析仪对致病菌进行鉴定,采用多重PCR方法分析致病菌CA26是否携带有溶血性毒素基因(eth A、eth B、esa C)。并对健康鲶鱼腹部注射致病菌CA26,采集侵染后6~96 h鲶鱼的血液和肝脏样品,使用分光光度计检测血液中的铁元素质量浓度,采用干法消化法检测鲶鱼肝脏中的铁元素质量浓度,实时荧光定量PCR检测铁元素调控基因的变化(以未侵染健康鲶鱼为对照)。结果表明,鲶鱼致病菌属于溶血性迟缓爱德华氏菌。与对照组鲶鱼相比,鲶鱼受到迟缓爱德华氏菌CA26侵染后,血液铁元素质量浓度降低,血液载铁蛋白质量浓度升高,肝脏铁元素质量浓度升高。肝脏铁调素基因(hepc)、白细胞介素6基因(il-6)、蛋白质酪氨酸激酶基因(jak3)、转录激活因子基因(stat3)表达量均升高。可见,健康鲶鱼受到迟缓爱德华氏菌CA26侵染后,hepc基因、il-6基因、jak3基因、stat3基因等表达量升高,从而降低鲶鱼血液铁元素质量浓度,增加血液载铁蛋白和肝脏铁元素质量浓度,有利于提高其机体免疫能力。

摘  要:
The pathogenic bacteria CA26 was isolated from Fengyang catfish liver samples by the method of plate streaking cultivation,identified by VITEK-32 automatic analyzer,and the carrying hemolytic toxin genes( eth A,eth B,esa C) were analyzed by multiple PCR. The effect of pathogen CA26 on the health catfish was studied at 6—96 h after infection,the concentration of iron in the blood was detected by the spectrophotometer,the concentration of iron in the liver was detected by the dry digestion method,and the changes of iron genes( health catfish as control) was detected by the real-time PCR. The results showed that the pathogenic bacteria of catfish belonged to the haemolytic Edwardsiella tarda. Compared with the control,the concentration of iron in blood decreased,the concentration of iron loaded ferritin increased,and the concentration of iron in liver increased after infected with Edwardsiella tarda CA26. The expression levels of liver hepcidin gene( hepc),interleukin 6 gene( il-6),protein tyrosine kinase gene( jak3)and transcription activator gene( stat3) increased. Therefore,the expression of hepc gene,il-6 gene,jak3 gene and stat3 gene increased after infection with Edwardsiella tarda CA26,thereby the concentration of blood iron decreased,the concentration of ferritin in blood increased,iron concentration in the liver increased,and the immune capacity improved.
