12种抗菌药物抗大肠埃希氏菌的增效剂筛选试验

摘  要:
筛选氟苯尼考等12种临床常用抗菌药物的抗大肠埃希氏菌增效剂。采用微量稀释法分别测定12种抗菌药物和丁香酚等8种可能成分的最小抑菌浓度(MIC);棋盘稀释法测定可能成分与抗菌药物联用抑菌浓度(FIC)指数,确定可能成分对抗菌药物的协同或相加、无关或拮抗作用。根据MIC和FIC指数分析发现,丁香酚、尼泊金酯钠、羧甲壳聚糖、没食子酸酯和乙基麦芽酚不仅具有抑菌作用且与多数抗菌药物联用对大肠埃希氏菌有明显的协同或相加作用,仅与少数抗菌药物联用存在无关或拮抗作用。丁香酚与庆大霉素等8种、尼泊金酯钠与金霉素等10种、羧甲壳聚糖与庆大霉素等11种、没食子酸酯与多西环素等9种、乙基麦芽酚与庆大霉素等12种抗菌药物具有协同或相加作用,可以作为抗大肠埃希氏菌类抗菌药物的抗菌增效剂候选药物,临床上联合使用可增加抗菌药物的疗效,降低抗菌药物在临床抗大肠埃希氏菌感染的用量。

摘  要:
To screen the anti-Escherichia coli synergistic agents of 12 antibiotic drugs such as florfenicol,minimal inhibitory concentration(MIC)of 12 antibacterial agents and 8 possible components including eugenol were determined by microdilution method.The checkerboard dilution method was used to determine the fractional inhibitory concentration(FIC)index of monomers and antibacterial agents to ascertain the synergistic or additive,irrelevant or antagonistic effects among them.MIC and FIC index analysis showed that the eugenol,sodium paraben esters,carboxychitosan,gallate and ethyl maltol not only had bacteriostatic effects,but also had synergistic or additive effects against E.coli when combined with the most of the12 antibacterial agents,while,only combined with a few antimicrobiols showed independent or antagonistic effects.Eugenol had synergistic or additive effects when it was combined with 8 antibacterial agents including gentamicin.So did sodium paraben combined with 10 antibacterial agents like chlortetracycline;carboxychitosan combined with 11 antibacterial agents including gentamicin;gallate combined with 9 antibacterial agents such as doxycycline;ethyl maltol combined with 12 antibacterial agents like gentamicin,which can be used as antibacterial synergistic agent candidate against E.coli.The combination of monomers and antibacterial agents can increase the efficacy as well as reduce the amount of antibacterial agents against E.coli infection in clinic.
