地黄多糖的超临界CO_2萃取及其对创伤小鼠脾脏的影响

摘  要:
为探讨地黄多糖对创伤小鼠脾脏和创面愈合的影响,采用超临界CO2萃取法提取地黄多糖,对创伤昆明种小白鼠灌服地黄多糖治疗,计算脾指数;石蜡包埋脾脏进行病理学分析;制备脾脏单细胞悬液,应用流式细胞仪进行检测,ModFit软件进行脾细胞周期分析。结果显示,地黄多糖的提取率达到30.4%;与对照组比较,模型组脾脏淋巴细胞稀疏,S期脾细胞比例和脾指数降低,而治疗组损伤程度减轻,S期、G2/M期脾细胞比例和脾指数升高,且创面愈合情况好。表明地黄多糖能够促进脾淋巴细胞的增殖,改善脾脏组织学变化,提高创伤小鼠脾指数和脾脏免疫功能,进而促进创面愈合。

摘  要:
To explore the effect of Rehmannia glutinosa polysaccharide on the spleen and wound healing in traumatized mice,Rehmannia glutinosa polysacchride was extractec by supercritical CO2 fluid.Rehmannia glutinosa polysacchride were administered once by intragastric administration(i.g.) to traumatized mice and then the spleen index was computed and the spleen was paraffin-embedded for pathological analysis.The FACSAria flow cytometry was used to detected single cell suspension and the spleen cell cycle was analyzed using ModFit software.The extraction rate of Rehmannia glutinosa polysacchride was 30.4%.Compared to the control group,spleen lymphocytes of the model group were scattered and the spleen index and S phase rate reduced.In treatment groups,the damage reduced but the spleen index,S phase and G2/M phase rate increased,and wound healing was better.This indicates that Rehmannia glutinosa polysacchride enhanced spleen index and spleen immune function in traumatized mice by promoting the proliferation of lymphocytes and spleen histology changing in traumatized mice,and thus to promote wound healing.
