菰黑粉菌中Ueubc2的克隆及表达分析

摘  要:
茭白是我国第二大水生蔬菜,它的形成与其内生真菌菰黑粉菌的侵染密切相关。根据菰黑粉菌转录组数据库设计特异引物,采用RT-PCR扩增获得Ueubc2基因,序列分析发现Ueubc2是玉米瘤黑粉菌中ubc2的同源基因,编码蛋白含有SAM、RA、SH3结构域,作用于MAPK信号通路。同时通过实时荧光定量PCR对Ueubc2基因进行表达分析,发现在融合生长过程中Ueubc2上调表达,并在菌丝形成时其表达量最大,随着菌丝生长表达量逐渐降低,表明其在菰黑粉菌真菌二型态转变过程中具有重要作用。

摘  要:
Zizania latifolia is the second aquatic vegetable in China. Its formation is closely releted the interaction which between its internally Endophytic fungus and Ustilago esculenta. According to the transcriptome database of Ustilago esculenta, we designed the specific primers, using RT-PRC to amplify gene Ueubc2. Sequence analysis showed that Ueubc2 is the homologous gene of ubc2 in Ustilago maydis, and the encoding protein contains SAM, RA and SH3 domain,which acting on the MAPK signal pathway. At the same time, the expression analysis by real-time PCR revealed to gene Ueubc2 was up-regulated during the mating progress, and reached highest when hyphae formed, then decreased during hyphae's growth, the result shows that it had an important role in the process of two type transformation of Ustilago esculenta.
