古尼拟青霉的种内杂交 Ⅰ.古尼拟青霉营养缺陷型原生质体的形成和再生

摘  要:
本文比较了不同酶液、渗透稳定剂、酶液pH、酶解温度、菌丝生长培养基成份及菌龄等因素对原生质体形成的作用及影响。用1.5%粗纤维素酶或1%脱壁酶、0.7MNaCl为渗透稳定剂,温度32℃、pH6、菌丝生长培养基为Sabouraud琼脂培养基,菌龄16小时,可以从所试的6株菌株的菌丝体得到大量的原生质体(4.0×10~6个/ml)。0.5MNaCl为稳定液,黄豆粉培养基上原生质体再生率可达11.4%。对原生质体释放和再生的形态学过程进行了描述和显微摄影。

摘  要:
The influencing factors on the formation of Protoplasts in P.gunnii 5-19, such as, stabilizers, temperatures, pH, age of mycelium, and media for mycelial growth were studied. When 0.7M NaC1 was used as osmotic stabilizer and Sabouraud agar medium as mycelial growth medium, at the temperature of 32℃, pH6, high yield of protoplasts was obtained from sixteen-hour mycelia of several auxotrophs by using 1% lywallzyme or 1.5% crude cellulase. When 0.5M NaC1 was used as stabilizer, about 11.4% of Protoplasts regenerated and formed colonies on soybean medium(SOS). The morphological processes of protoplasts formation and regeneration were illustrated by microPhotograph.
