荧光探针法研究茶多酚与DNA的相互作用

摘  要:
利 用荧光探 针 E B(溴 化乙啶)研 究茶多酚 ( T P)与 D N A 的相 互作用 及顺 铂( C D D P)对其 的影响,结果表明:在 E B D N A 体系中加 入 T P 后,荧光光谱 形状和峰位几乎 无变化,而荧光强度 明显下降; T P 压低 E B D N A 荧 光强度的效率 D 与其浓度成正相关,高浓度 T P 可以完全抑制 E B D N A 荧光强度⒚ T P与 C D D P 对 E B D N A 荧光压低作用存在叠加现象,表明 T P 可增强 C D D P 的药用效果,又 可降低其副作用⒚ T P使 E B D N A 体系的荧 光偏振度 P增加,而荧光寿命 τ缩短,这表明 T P 与 D N A 作用的机理可能是 T P 插 入了 D N A 分子碱 基对平 面中,影 响了 D N A 分子双 链的有 序度,并 使从 D N A 分子中 向 E B 分子传递的能量减少⒚

摘  要:
The authors made studies on the interaction between TP and DNA, and the effects of CDDP (cisplatin) on it by using fluorescence EB probe (Ethidium Bromide). The results showed that after adding TP to EB DNA system, the shape and the peck site of fluorescence spectrum no change, but fluorescence intensity decreased obviously The decreasing efficiency (D) was positive with TP concentration High concentration of TP could totally control the fluorescence intensity of EB DNA TP and CDDP had an cooperative effect on DNA, which indicated TP could not only increase CDDP's positive medical effect, but also decrease its side effect TP increased fluorescence polarization of EB DNA and slightly shortened fluorescence lifetime (τ) These indicated that the mechanism of interaction between TP and DNA was perhaps that TP inserted into the base pairs plane of DNA, which affected the sequency of twin stranded DNA and decreased the energy transmitted from DNA to EB These results showed the anti cancer mechansim of TP from another point of view That is, TP maybe could prevent cancer by affecting the structure and function of DNA
