牛磺鹅去氧胆酸对佐剂性关节炎大鼠成纤维样滑膜细胞ICAM-1表达的影响

摘  要:
为了研究牛磺鹅去氧胆酸(taurochenodeoxycholic acid,TCDCA)对佐剂性关节炎模型大鼠成纤维样滑膜细胞(fibroblast-like synoviocytes,FLS)中细胞间黏附分子-1(intercellular cell adhesion molecule-1,ICAM-1)表达的影响,采用qPCR与ELISA法检测了FLS中ICAM-1的表达量,结果表明,1×10~(-6)~1×10~(-4) mol/L TCDCA在基因与蛋白水平均能够显著或极显著抑制IL-1β刺激下FLS中ICAM-1的表达(P<0.05或P<0.01),且该抑制作用可被糖皮质激素受体(glucocorticoid receptor,GR)阻断剂米非司酮(RU486)拮抗,提示TCDCA对佐剂性关节炎的治疗作用与抑制GR活性进而抑制ICAM-1表达有关。

摘  要:
To investigate the effect of taurochenodeoxycholic acid on the expression of intercellular cell adhesion molecule-1(ICAM-1)in fibroblast-like synoviocytes(FLS)of adjuvant arthritis(AA)in rats,qPCR and ELISA were used to measure the mRNA and protein expression of ICAM-1,respectively.The results showed that the gene and protein expression of ICAM-1were inhibited significantly by TCDCA at the concentration of 1×10~(-6)-1×10~(-4) mol/L,and the inhibition of TCDCA could be blocked by glucocorticoid receptor(GR)inhibitor mifepristone(RU486),which suggests that the mechanism of TCDCA treatment for AA was related to inhibition of ICAM-1 expression mediated by GR.
