多肋藻渣中褐藻胶提取工艺及其分离纯化研究

摘  要:
以多肋藻加工副产物——藻渣为原料,采用碱提法探讨多肋藻渣中褐藻胶的最佳提取工艺参数,并进一步分离纯化。正交实验结果表明,在提取时间4.5h、料液比1∶40、碳酸钠浓度3.5%、温度70℃时,多肋藻渣中褐藻胶的粗得率为44.07%,提取物纯度为94.77%,粘度为14.8m Pa·s(1.0%样液)。褐藻胶经D301-T弱碱性阴离子树脂交换层析分级得到F1、F2两个组分,其回收率分别为19.33%和63.33%;褐藻胶含量分别为84.56%和89.69%。将组分F1、F2分别经Sephacryl S-300凝胶柱层析纯化后均得到一个单一组分,其相对分子质量分别为257、81ku。

摘  要:
The extraction of alginate from Costaria costata residues was studied by alkali test,then it was further isolated and purified.The optimum extraction condition by orthogonal test was as follows: 4.5 hours extraction time,1∶ 40 ratio of solid to liquid,3.5% Na2CO3 concentration and 70℃ extraction temperature,the crude yield was44.07%,with 94.77% content of sodium alginate.The viscosity was 14.8m Pa·s. Sodium alginate was further purified by D301- T weak- base exchange anion resin,two fractions( F1 and F2) were obtained with the recovery yield of19.33% and 63.33%,which sodium alginate content of 84.56% and 89.69%,respectively. F1 and F2 were then separated by Sephacryl S-300 gel chrom atography,then the molecular weights of F1,F2 were estimated as 257 ku and 81 ku,respectively.
