多肋藻岩藻聚糖硫酸酯的提取及其降血脂作用研究

摘  要:
通过对多肋藻Costaria costata基本成分的测定与对多肋藻中活性成分岩藻聚糖硫酸酯提取工艺进行优化,研究了提取的岩藻聚糖硫酸酯对小鼠血清高血脂的抑制作用。结果表明:多肋藻的水分、灰分、蛋白质、脂肪含量分别为10.55%、15.40%、11.13%、2.00%(质量分数,下同),多肋藻的总膳食纤维、碘、甘露醇含量分别为48.05%、0.10%、11.30%。正交试验结果表明,岩藻聚糖硫酸酯的优化提取工艺为酶加量0.10%、温度50℃、pH 5.5、酶解时间30 min,在此条件下岩藻聚糖硫酸酯的提取率为1.859%,总糖与总硫酸根含量分别为1.416%和0.972%。建立高血脂症小鼠模型,采用岩藻聚糖硫酸酯粗提物对其灌胃,试验结果表明:各剂量组对小鼠血清总胆固醇(TC)均具有显著抑制作用(P<0.05);剂量高于500 mg/(kg.d)时,对小鼠血清甘油三酯(TG)和低密度脂蛋白(LDL-C)具有显著抑制作用(P<0.05)。多肋藻岩藻聚糖硫酸酯粗提物对高血脂症中的三项指标均具显著抑制作用,表明其有一定的降血脂功能。

摘  要:
The approximate composition of seaweed Costaria costata was analyzed,the extraction of fucoidan was optimized from the sea weed and the inhibition of high serum lipid in mice by the fucoidan was studied.It was found that the Costaria costata contained 10.55% moisture,15.40% ash,11.13% protein,2.00% fat(mass percent,et sequentia),48.05% total dietary fiber,0.10% iodine,and 11.30% mannitol.The orthogonal test showed that the best extraction of fucoidan was observed under the conditions of enzyme addition of 0.10%,temperature 50 ℃,pH 5.5,and hydrolysis period 30 min with the extraction rate of 1.859%,the total sugar of 1.416% and sulfate radicals of 0.972%.The hyperlipemia mice by intragastrical injection of the crude fucoidan extract showed that significant inhibition of serum total cholesterol(TC)(P<0.05),specifically inhibition of the mice serum triglyceride(TG) and low density lipoprotein(LDL-C) at a dose of over 500 mg/(kg·d)(P<0.05).The fucoidan extract from the Costaria costata had significant inhibition of the three indices of hyperlipidemia mentioned obove,indicating that the fucoidan functions as high-lipid reducing.
