巴西橡胶树REF/SRPP家族基因的克隆及表达分析

摘  要:
巴西橡胶树(Hevea brasiliensis)胶乳是天然橡胶的主要来源,橡胶延伸因子(REF)和小橡胶粒子膜蛋白(SRPP)作为胶乳中的两大主要蛋白,被认为参与了橡胶的合成和调控。目前,NCBI蛋白数据库已经登录了巴西橡胶树REF/SRPP家族的6个成员,我们通过研究又获得8个新的REF/SRPP基因家族成员,Hb REF154、Hb REF221、Hb REF296、Hb REF542、Hb SRPP178、Hb SRPP203、Hb SRPP216、Hb SRPP234。这14个成员编码蛋白都具有保守的REF结构域,有两个成员还具有ATP合酶亚基结构域,一个成员具有甲基结合结构域。进化分析显示橡胶树REF/SRPP家族成员与产胶植物REF/SRPP家族成员在进化上较为接近。q PCR分析发现该家族成员都响应割胶刺激表达升高,除Hb REF138外其它成员都响应乙烯处理表达降低。通过shotgun数据分析发现,除Hb REF221外的13个成员都存在于橡胶粒子或C-乳清组分中,Hb REF138、Hb REF154、Hb REF175和Hb REF258主要存在于橡胶粒子中,Hb REF542、Hb SRPP216是橡胶粒子中所特有的,Hb REF296和Hb SRPP200是C乳清中所特有的。该研究结果揭示了REF/SRPP家族成员及其基本的表达特性,为深入研究该家族成员在巴西橡胶树产胶中的具体作用提供了一定的基础。

摘  要:
The latex of rubber tree(Hevea brasiliensis) is the major source of nature rubber production. The rubber elongation factor(REF) and small rubber particle protein(SRPP) are the two major proteins in the latex, and they were honowns as involve in the regulation of natural rubber biosynthesis. At present, six REF/SRPP members have been identified in Hevea, and registered in NCBI protein database. In our study, 8 new REF/SRPP gene members were obtained from the transcriptome data and homological cloning, and they were named as Hb REF154,Hb REF221, Hb REF 296, Hb REF542, Hb SRPP178, Hb SRPP203, Hb SRPP216, and Hb SRPP234 based on the number of amino acids of the protein. All the proteins of the 14 members have the REF conservative domain of the family, two of them also have an ATP synthesis subunit domains, and one of them has a methyl-binding domain.Phylogenetic analysis shows that REF/SRPP members in Hevea are close to REF/SRPP members in latex plants instead of congeneric plant. q PCR analysis discovered that the expression of all the 14 genes were promoted by tapping and decreased by ethylene stimulation, excepted Hb REF138. Shotgun data analysis discovered that 13 members except Hb REF221 were exist in the proteins of rubber particles or C-serum faction of latex, among of them, Hb REF138, Hb REF154, Hb REF175 and Hb REF258 were mainly distributed in rubber particles, Hb REF542 and Hb SRPP216 were only exist in rubber particles, and Hb REF296 and Hb SRPP200 only exist in C-serum. The studies discovered the members of REF/SRPP in rubber tree(Hevea brasiliensis), exhibited the expression pattern of them response to tapping and ethylene stimulation and the distribution of them in the latex. The results take some helps for the studies of specific roles of REF/SRPP family members in latex production of rubber tree(Hevea brasiliensis).
