陕北木枣基因组DNA提取方法研究

摘  要:
采用SDS法和改良CTAB法对不同方式保存的陕北木枣叶片基因组DNA进行了提取,并对木枣不同取材部位(叶芽,花蕾,花瓣,嫩茎,韧皮部,嫩叶)的基因组DNA提取效果进行了研究。结果表明:改良CTAB法提取的经-80℃快速冷冻的叶芽或幼嫩叶片获得的结果相对最好,即得到的DNA蛋白质等杂质去除较彻底,条带较整齐,产量较高。

摘  要:
Using SDS method and improved CTAB method,total DNA was extracted from leaves of Muzao Jujube from Northern Shanxi with different ways to preserve.Moreover,the efficient of extracting total DNA from different parts of Muzao jujube(leaf buds,flower buds,petal,ligneous phloem,tender stems and young leaves) were compared.The results showed that the total DNA extracted by improved CTAB method was the best when materials derived from leaf buds or young leaves stored at-80℃,namely the performance for protein impurities interference less,DNA structure full and purity and productivity high.
