环孢素A对咖啡因刺激下的L6细胞钙蛋白酶活性的影响

摘  要:
探究钙调神经磷酸酶抑制剂环孢素A是否调节骨骼肌细胞钙蛋白酶活性。将分化8 d后的L6肌管细胞随机分为4组,即对照组、环孢素组、咖啡因组、环孢素+咖啡因组。急性刺激5 h后收集细胞,提取并定量细胞总蛋白和肌原纤维蛋白,用荧光定量法测定钙蛋白酶的活性。结果显示,咖啡因刺激后肌原纤维蛋白含量下降,同时钙蛋白酶活性明显增加;加用环孢素则显著增加咖啡因刺激时的肌原纤维蛋白含量,同时咖啡因+环孢素组钙蛋白酶活性明显低于咖啡因刺激组。因此,环孢素A抑制咖啡因刺激的L6细胞钙蛋白酶活性。

摘  要:
To investigate whether calcineurin regulates calpain activity in skeletal muscle cells.L6 myotubes differentiated for 8 days were divided into four groups: control group;cyclosporine group;caffeine group;CsA + caffeine group.Stimulated the cell and collected them after 5 h,extrccted and quantified the total protein and myofibril protein.The activity of calpain was measured by fluorescence quantitative assay.The results showed that myofibril protein content was decreased with concomitant an increased calpain activity after caffeine stimulation.Compared with caffeine group,caffeine + CsA group markedly enhanced protein content with decreased calpain activity.Hence,CsA inhibits caffeine-stimulated calpain activity in L6 cells.
